Protocol for intravitreal injection
Intravitreous injection is a widely used technique in visual sciences research. It can be used to establish animal models with ocular diseases or as direct application of local treatment. We present here a protocol that uses a intravitreous injection with a viral vector in the mouse. The experimental process of intravitreous injection is as follows.
STEP 1
Before surgery, mouse is anesthetized by intraperitoneal injection of 2 % sodium pentobarbital (50 mg/kg). Wait until the mouse becomes motionless (~5 min).
STEP 2
Apply eye ointment such as Oculentum simplex, Teva Pharmachemie to the mouse eyes as to keep eyes moist.
STEP 3
Position the mouse and expose the superior nasal region of the eye, then use a 30-gauge needle to puncture the superior nasal sclera at the level of the par plana and gently inject the virus. when finishing injection, keep the needle in place for about a few seconds, then gently remove it. In general, the injection volume is 1~3 μl/eye. Please note that avoid touching the ocular muscle and vessels.
STEP 4
Generally, each mouse is injected with only one eye, and the other one serves as a control. Topical antibacterial treatment on virus-injected eyes is necessary to prevent infection.
STEP 5
After one week, use a fundus camera for fundus examination.
